I just bought a used 2006 wee with 8500 miles. The TBS has not been done. The manual says to do the TBS at 7500 miles. The bike seems to run OK. Is this something that should be done now, or only if the bike starts running rough or other symptoms?

Impossible to say, but probably not. Still, it's not too hard to check it with a homemade manometer; the hard part is getting the hoses onto the nipples on the TBs. If you do it, I highly recommend that you permanently attach two lengths of vacuum hose to each nipple and run them out to locations where they can be easily accessed for future TB checks/synchs.

The plugs are good for well over 20,000 miles. Don't bother. Wait until it's time for a valve clearance check when they are easily accessed.
